The experimental results showed that the optimal conditions were 40% tert-amyl alcohol in mixed solvent, molar ratio of fructose to palmitic acid 1 ∶ 2, some 4A molecular sieve, 60 ℃, rock bed rotate speed 180 r/ min. 实验结果表明,较好的合成条件为果糖与棕榈酸的摩尔比1∶2,叔戊醇占混合溶剂体积总量的40%,0.1g酶,4A型分子筛适量,反应温度60℃,摇床转速180r/min。

In this work, to enhance the thermal stability of immobilized enzyme, small molecule reagents such as alanine, valine, tryptophan, tert-amyl alcohol were used to quench and block excessive activated groups on support surface. 因此本文加入小分子物质封闭载体表面的多余基团,阻止其对固定化酶的迫害。丙氨酸、缬氨酸、色氨酸、叔戊醇等小分子都大大提高了固定化嗜热菌蛋白酶的热稳定性。
